Mini Lab Report 2

Reports must be typed and submitted to Canvas by the start of your next lab (see the “Course Summary” tab for links to all assignment submissions). Lab notes are not part of the mini lab reports and should not be submitted.

This mini lab report should include methods, results and discussion sections for experiments in labs 5-7.

Methods Section:

  • Report methods used in YAC-assembly reaction and transformation in lab 5, picking yeast colonies and the multiplex PCR reaction in lab 6 and gel electrophoresis in lab 7.
  • Be sure to include:
    • Purpose statement/ brief summary of each experiment
    • The amount of DNA used and reagents used in the YAC-assembly reaction
    • Transformation/heat shock: key information in the procedures, such as temperature and time
    • Yeast plate incubation and colony picking
    • Colony DNA processing and PCR reaction assembly
    • Gel preparation, running and imaging
  • A few general reminders:
    • Your method section should be clear enough that another scientist should be able to repeat your experiments.
    • Some details can be omitted, such as getting ice or other minor details.

Results Section:

  • Report results of the transformation (plate colony counts) and the gel image.
  • Be sure to include:
    • Purpose statement/ brief summary of each experiment.
    • Yeast plates:
      • How many colonies from each plate? Describe the difference.
      • Include a picture of the plates (with figure legend).
    • PCR results:
      • How many bands in each lane? What are their sizes?
      • Are all colonies have bands? Describe any difference.
      • Include a picture of the gel: label each lane; highlight important lanes if preferred; label most if not all of the size marker; figure legend
    • If you don’t have colonies:
      • State that you did not obtain any colonies – do not report data from another student’s plate or instructor’s plate.
      • Include a picture of your plates(with figure legend).
    • Be careful not to add discussion text here! Results section should include what you observed, not why you think you observed it.

Discussion Section:

  • Include the following information:
    • Recap what your results were in one or two sentences.
    • Discuss possible reason of any differences in colony numbers on the two plates.
    • If you have no colonies, you should include a statement on why you may have found no colonies and how to improve.
    • Discuss the number of bands in your agarose gel.
    • Discuss any difference in the number or size of the bands in different yeast colonies.
    • Discuss which or any of the colonies may have the correct product and your reasoning.
