Lab 5: Screening for pETblue2-HCAII clones

Determine whether the HCAII gene is present in pETblue2 in each of your potential clones; by:

• Using the blue/white screening technique to identify bacteria suspected to contain your clone of interest
• Purifying DNA from these potential clones
• Digesting the purified DNA with restriction enzymes to differentiate positive vs. negative clones (i.e. a restriction digest screen)

As most of the techniques you will use this week have already been used in previous labs, you will have the opportunity to work more independently.
