Lab 9: PRELAB
PRE-LAB ASSIGNMENT
- What will you be doing in lab? Why?
- Take another look at the tryptophans in HCAII that you studied in Lab 1. List the tryptophans here and whether you decided they were completely buried, minimally exposed, significantly exposed, or fully exposed. Does the mutation you are studying destroy or create a tryptophan? If so, how might the mutation itself affect this experiment? Is the mutation you are studying very close to a tryptophan residue? How might this affect the experiment?
- Be sure you know the concentration of your protein sample (in μM, calculated using Beer’s law).
- Complete planning and calculations for the lab PRIOR to lab!
- You must coordinate with your podmates
- Using the Google Sheets template document as a starting point: Lab 9 Planning Template
- This video from a TA helps explain the planning process:
PRE-LAB QUIZ
Please answer the following true/false questions:
- A mutation that affects protein stability should also affect the CM
- A decrease in CM is likely indicative of a decrease in protein stability
- A decrease in ΔGunfolding indicates an increase in protein stability
