Lab 5: Screening for pETblue2-HCAII clones
Lab 5 Objectives
Determine whether the HCAII gene is present in pETblue2 in each of your potential clones; by:
- Using the blue/white screening technique to identify bacteria suspected to contain your clone of interest
- Purifying DNA from these potential clones
- Digesting the purified DNA with restriction enzymes to differentiate positive vs. negative clones (i.e. a restriction digest screen)
