Lab 10: Investigation of the wild type and mutant HCAII enzyme activity
Lab 10 Objectives
- Gain hands-on experience with enzyme kinetics
- Use an enzyme assay to determine kcat, KM, and catalytic efficiency of wild type and mutant HCAII esterase activity
- Interpret the results of this assay to determine whether your mutation affects catalytic activity of HCAII
Before lab, you will complete a short at-home experiment on enzyme kinetics from Carolina Distance Learning. In lab, you and your partner will discuss this experiment, then work on the HCAII experiment.
